Drifter Cole Harden is accused of stealing a horse and faces hanging by self-appointed Judge Roy Bean, but Harden manages to talk his way out of it by claiming to be a friend of stage star Lillie Langtry, with whom the judge is obsessed, even though he has never met her. Tensions rise when Harden comes to the defense of a group of struggling homesteaders who Judge Bean is trying to drive away.

Two investigators for a stagecoach company are assigned to find out why the company's stages keep being ambushed. They discover that the culprits are white men disguised as Indians, and they set out to discover who is behind the plot.

To investigate a gold-shipping scam, a postal inspector goes undercover and tries to infiltrate the gang he believes is responsible.

McGowan). Davidson allows Slade to escape from jail and follows him to aid him in proving his innocence.

The notorious outlaw Black Bart has reappeared and the Range Busters are sent to investigate. When they find that Black Bart is now a respectable citizen and that someone is impersonating him, they set a trap for the robber.

Hammond is after the Craig ranch and has framed Charlie Craig for murder. Mother Craig brings in the Range Busters. They capture one of Hammond's men and Alibi plans to trick him into a confession as to who the real murderer is. Meanwhile, Denny has overheard Hammond's plans for his next move and he and Crash set out to round up the gang.

Ace Remsen and Deuce Remsen are twin brothers. Deuce is a bandit and he is slain by Ace. The good Ace goes undercover as Deuce in order to solve the murder of the rancher Seth Waverly. During his investigation Ace falls in love with Seth's attractive daughter Merrill.

The Beagles and the Hickses are two mountain families that have been feuding all their lives. The Hickses come up with a plan to get rid of their enemies once and for all by hiring two sharpshooters to finish them off. Turns out that the "sharpshooters" aren't quite all they're cracked up to be, resulting in some unintended consequences.

Roy is a newspaper reporter. He goes to Cheyenne to cover the activities of supposed bad guy Arapahoe Brown. Roy, of course, discovers who the real bad guy is.

Crenshaw and Randolph are competing freight haulers and Randolph's lead man Tom Baxter has given him an advantage....

Dorothy, and her big city lawyer boyfriend, return to the Lazy 'B' ranch to read her late father's will. For Dorothy to inherit everything, she must stay on the ranch for 5 years. If she does not, everything goes to Buck, who is the manager. She does not like Buck, so she makes a deal with the wrong people for cattle and then the outlaws go to the ranch to get the $10,000 from her. But Buck is on the job.

Jack Burton saves Anita Leland from a runaway horse, and then saves Sam Sills from a beating at the hands of Bill Sterns. Jack goes into partnership with Sam, who is later murdered by Bill. Jack is arrested for the murder on circumstantial evidence. He escapes and finds Bill just in time to save Anita from his foul advances. Bill is arrested, while Jack and Anita anticipate a happy future together.

Bob Allen in his starring debut gets a job on Wright's ranch where he hopes to find the rustlers no one else has been able to locate. Everyone is looking for men when the actual rustler is a horse.

Johnny Mack Brown's Universal western series was drawing to a close when Cheyenne Roundup was released in mid-1943. Brown is herein cast in a dual role, as honest Gils Brandon and his less-than-honest brother Buck. Pursued by lawman Steve Rawlins (Tex Ritter), Buck tries to pass himself off as the upright Gils.

Ranger Ray plans to marry stage driver Bill Mason's daughter Mary, but there are problems ahead....

Mortimer builds a fence for the cattle brought by Ken Morley. To retaliate, Slater who wants access to the land, builds a dam cutting off Mortimer's water supply. When Ken confronts Slater, he is captured. Then lightning destroys the dam and Ken, imprisoned in a shack, is in the path of the oncoming water.
